GCMS and LCMSMS In Residue Analyses with instruments, many critical parameters, from the cleanliness of the instruments to LOD values, are directly linked to the sample preparation procedure. In 2003 USDA one of the researchers Lehotay and colleagues developed by QuEChERS (Quick, Easy, Cheap, Effective, Rugged and SThe practical sample preparation procedure gained by laboratories with the afe) method is a particularly widely used AOAC 2007.01 method and started to be used in all laboratories.

Why QuEChERS?

  • Since much less reagent is used, it provides clean extraction and reduces polar interferences.
  • It is advantageous because it does not use Acetic Acid which may cause problems in GC/MS analyses.
  • It is 4 times faster than other methods.
  • It consumes 6-9 times less solvent than other methods.
  • Does not create chlorinated waste, more sensitive to the environment.

Why AOAC 2007.01

- Prevents degradation of base sensitive analytes by using %1 acetic acid and sodium acetate buffer in acetonitrile. It makes it possible to analyse fruits with high water content and high acid content as a single matrix (according to Sanco Directive).

- According to USDA studies, it provides much better recovery of pH sensitive compounds compared to other QuEChERS methods.

- Acetic acid used in the extraction step is inactivated with the PSA sorbent used in the Clean-Up step, eliminating possible GC resolution problems.

Clean-Up Reagents and Intended Use

Endcapped C18 It provides removal of long chain fatty compounds, sterols and non-polar interferences.
Graphitised Carbon Black (GCB) It is used for the removal of pigments, polyphenols and other polar compounds; it can cause the removal of polar aromatic pesticides such as Chlorothalonil, Coumaphos, Hexachlorobenzene, Thiabendazole, Terbufos and Quintozene.
MgSO4 Removes water from the organic phase.
Primary Secondary Amine (PSA) Removes sugars, fatty acids, organic acids, oils and some pigments. It can remove more fat and sterol when used with C18.
